Description of SB 265610

SB 265610 is a competitive non-peptide allosteric CXCR2 antagonist that blocks neutrophil chemotaxis (IC50=70nM) and calcium mobilization (IC50=3.7nM). SB 265610 can be used for research related to inflammatory diseases and cancer[1-4].

In vitro, SB 265610 (1–20μM) was used to treat SW480 cells for 48 hours. SB 265610 failed to induce G2-M phase arrest[5]. SB 265610 (1μM) was used to treat PyMT-Luc and 17L3C-Luc breast cancer cell lines for 4 hours. SB 265610 significantly attenuated mesenchymal stem cell-conditioned medium-induced cancer cell migration and reduced CXCR2 phosphorylation[6].

In vivo, SB 265610 (2mg/kg; once daily) was intraperitoneally injected into BALB/c or C57BL/6 mice bearing MC38 or CT26 colorectal cancer cells for 12 days. SB 265610 blocked the tumor-promoting effect of METTL3[7]. SB 265610 (100mg/kg/day) was administered to Nitrogen mustard-induced chemical burn model C57-Black mice via dietary supplementation for 5 days. SB 265610 significantly lowered myeloperoxidase activity and impaired the wound healing process[8].

SW480 cells (colorectal carcinoma cell line)

Preparation Method

SW480 cells were maintained in DMEM supplemented with 10% fetal bovine serum (FBS), L-glutamine, penicillin, and streptomycin at 37°C in a humidified atmosphere of 5% CO₂. Cells was treated with SB 265610 at concentrations of 1, 10, and 20μM for 48 hours.
