Sample solution is provided at 25 µL, 10mM.
T4 Polynucleotide Kinase, abbreviated as T4 PNK, is a polynucleotide 5 'hydroxy kinase that catalyzes ATP γ The phosphate group is transferred to the 5 'hydroxyl group of single or double stranded DNA, RNA, oligonucleotides, or single nucleotides with a 3' phosphate group. Other NTPs can also produce the same reaction: 5 '- OH+NTP → 5' - P+NDP.
Operation process
Reaction system setup:
1) Dissolve and mix all solutions required for PCR reaction. Place on ice bath or in ice box. It is recommended to use PCR liquid in aliquots to avoid repeated freezing and thawing.
2) Refer to the reaction set up in the table below. It is recommended that the PCR reaction system be configured in an ice bath or on an ice box:
| Reagents | 50 μl reaction system |
| T4 Polynucleotide Kinase (10 U/ul) | 1-2 μl |
| 5×T4 PNK Buffer | 10 μl |
| DNA to be phosphorylated | — |
| γ-[32P]-ATP/ATP(10mM) | 5 μl |
| T4PNK, template, ATP can be adjusted according to the specific amount of the experiment, less than 50 μl can be supplemented with RNaseFree-H2O | |
| Volume | 50 μl |
3) Incubate at 37°C for 30 minutes.
| Components | 100U | 500U |
| T4 Polynucleotide Kinase(10 U/ul) | 10 µl | 50 µl |
| 5X T4 PNK Buffer | 100 µl | 500 µl |
