Description of TRF

Transferrin is the main iron transfer protein in plasma, which provides iron for cell differentiation and cell metabolism. It was found that all cells can not grow without transferrin. Not only does maintaining normal cell metabolism depend on iron in bioactive form, iron is also a cofactor of some enzymes, such as RNA polymerase and DNA synthase. It is also an important part of hemoglobin.

Transferrin is a plasma glycoprotein isolated from plasma. Bovine transferrin exists in two molecular forms with molecular weights of 74kd and 78kd respectively. Each transferrin molecule can bind two molecules of iron ions through the C-terminal and N-terminal structural sites, so transferrin exists in three forms: de iron (not bound to iron ions), saturated iron (bound to two molecules of iron ions) and partial saturation. Under physiological conditions, only 1 / 3 of transferrin molecules are saturated by iron.

Instructions: transferrin is necessary for most cell lines to grow in serum-free medium. Studies have shown that transferrin in saturated iron form is the most effective form to promote cell growth. For hybridoma, adherent cells and suspension cell lines, the concentration of transferrin in the culture medium should be kept at 0.5-100 during cell culture μ g/ml. For different cell lines, the most appropriate concentration needs to be selected.

Transferrin lyophilized powder needs to be stored in cold storage and dissolved before use. Generally, the transferrin lyophilized powder is dissolved into storage solution with PBS or other suitable solvent at room temperature, with the concentration of 0.5-2.0mg/ml, and then filtered with sterile filter membrane. The filtrate can be stored at 2-8 ℃ under sterile conditions for 30 days.

In vivo Formulation Calculator (Clear solution) of TRF

Step 1: Enter information below (Recommended: An additional animal making an allowance for loss during the experiment)

mg/kg g μL

Step 2: Enter the in vivo formulation (This is only the calculator, not formulation. Please contact us first if there is no in vivo formulation at the solubility Section.)

% DMSO % % Tween 80 % saline
%DMSO %

Calculation results:

Working concentration: mg/ml;

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL saline, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such as vortex, ultrasound or hot water bath can be used to aid dissolving.
3. All of the above co-solvents are available for purchase on the GlpBio website.
