Description of BODIPY 493/503 methyl bromide

BODIPY 493/503 methyl bromide is a fluorescent fatty acid analog with maximum excitation/emission wavelengths at 493/503nm[1]. BODIPY dye is a small molecule dye with strong ultraviolet absorption ability. Its fluorescence peak is relatively sharp and its quantum yield is high. It is relatively insensitive to the polarity and pH value of the environment and is relatively stable under different physiological conditions[2]. Due to its asymmetric structure, BODIPY has a variety of derivative structural products[3]. BODIPY lipid droplet dyes can easily pass through the cell membrane and enter the cell interior. They can be specifically stained by localizing on neutral lipids in the cell, so the dye can be used to label living cells and fixed cells[4, 5].

Protocol of BODIPY 493/503 methyl bromide

This protocol is only a guide. Please modify it according to your specific needs.

1. Preparation of staining solution

(1) Stock solution: Dissolve BODIPY 493/503 methyl bromide in DMSO to prepare a stock solution with a concentration of 1-10mM.

Note: After unused stock solution is aliquoted, store it at -20°C or -80°C in the dark to avoid repeated freezing and thawing.

(2) Working solution: Dilute the stock solution with a suitable buffer (such as serum-free medium or PBS) to prepare a working solution with a concentration of 1-10μM.

Note: Please adjust the concentration of the working solution according to the actual situation and prepare it before use.

2. Cell staining (suspended cells)

(1) Collect cells by centrifugation and wash twice with PBS, each time for 5min.

(2) Add 1mL of BODIPY 493/503 methyl bromide working solution to resuspend the cells. The recommended cell density is 1×106/mL, incubate at room temperature in the dark for 5-30min. The optimal incubation time for different cells is different. Please explore it yourself according to the specific experimental needs.

(3) After the incubation is completed, centrifuge at 400g for 3-4min, remove the supernatant, and add PBS to wash 2-3 times, 5min each time.

(4) After resuspending the cells with 1mL of serum-free culture medium or PBS, use a fluorescence microscope or flow cytometer to observe.

3. Cell staining (adherent cells)

(1) Culture adherent cells on sterile coverslips.

(2) Remove the coverslip from the culture medium and aspirate the excess culture medium.

(3) Add 100μL of dye working solution from one corner of the coverslip and gently shake to evenly cover all cells with the dye.

(4) Incubate at room temperature in the dark for 5-30min. The optimal incubation time for different cells is different. Please explore it yourself according to the specific experimental needs.

(5) After the incubation is completed, aspirate the dye working solution, wash the coverslip with culture medium 2-3 times, and use a fluorescence microscope or flow cytometer to observe.

Notes:

(1) It is recommended to set up a positive control. The cells in the control group are incubated with 30μM oleic acid for 8h before subsequent experiments;

(2) All fluorescent dyes have quenching problems. Please try to avoid light to slow down fluorescence quenching;

(3) For your safety and health, please wear a lab coat and disposable gloves during operation.
