Rigin is an N-terminally palmitoylated tetrapeptide skincare active ingredient. Rigin can be used for research related to skin inflammaging, photoaging, and anti-wrinkle cosmetic ingredients[1-4].
In vitro, Rigin (2.2μM) was co-administered with palmitoyl oligopeptide (4.7μM) to human dermal fibroblasts for 3 days. Rigin promoted the synthesis of collagen I, fibronectin, and hyaluronan, and upregulated the expression of procollagen, lysyl oxidase, fibronectin, laminin, and other related genes[5].
In vivo, Rigin (3% or 5% gel; 100μL per application) was topically applied to PM10-treated SKH-1 hairless mice (the dorsal skin of 1cm² was first treated with 10μL of PM10 suspension; the peptide gel was applied daily) for 4 consecutive weeks. Rigin reduced PM10-induced expression of IL-1β and IL-6 in the skin[6].

Preparation Method | The dorsal skin (1cm²) of hairless mice was topically applied with PM10 suspension (10μL) to induce cutaneous inflammatory response. Subsequently, 3% or 5% Rigin (100μL per application) was topically applied to the same site. After treatment, skin tissues were collected for immunohistochemistry and Western blot analysis to evaluate IL-1β and IL-6 expression levels. |
