Description of Peginterferon alfa-2a

Peginterferon alfa-2a (Ro 25-8310/000) is a 40 kDa branched pegylated IFN alfa-2a with antiviral and immunomodulatory activities. Peginterferon alfa-2a binds to the Interferon Receptor on the surface of target cells, induces the formation of ISGF3, and promotes the expression of effector proteins such as interferon-stimulated genes (ISGs) and Mx proteins, thereby exerting sustained antiviral effects. Peginterferon alfa-2a is applicable to research on virus infections associated with chronic hepatitis C and chronic hepatitis B [1] [2].

In Vivo, Peginterferon alfa-2a (Ro 25-8310/000) (40 kDa; subcutaneous injection) extends the terminal elimination half-life to 15.0 h and the mean plasma residence time to 20.0 h in rats, and produces more sustained plasma exposure compared with conventional IFN alfa-2a[1]. Peginterferon alfa-2a (40 kDa) distributes to the liver and kidneys in rats. Intact Peginterferon alfa-2a remains detectable in the blood and liver for at least 24 h after administration, and exhibits reduced renal clearance and prolonged hepatic antiviral activity[1].

In Vitro, Peginterferon alfa-2a (Ro 25-8310/000) promotes the formation of ISGF3 by binding to Interferon Receptor on the surface of target cells, and induces the expression of antiviral effector proteins such as ISGs and Mx proteins, thereby exerting antiviral and immunomodulatory effects[2]

Calculate

In vivo Formulation Calculator (Clear solution) of Peginterferon alfa-2a

Step 1: Enter information below (Recommended: An additional animal making an allowance for loss during the experiment)

mg/kg g μL

Step 2: Enter the in vivo formulation (This is only the calculator, not formulation. Please contact us first if there is no in vivo formulation at the solubility Section.)

% DMSO % % Tween 80 % saline
%DMSO %

Calculation results:

Working concentration: mg/ml;

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL saline, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such as vortex, ultrasound or hot water bath can be used to aid dissolving.
3. All of the above co-solvents are available for purchase on the GlpBio website.
