Rozanolixizumab (UCB7665), a humanized high-affinity anti-human neonatal Fc receptor (FcRn) monoclonal antibody (IgG4P), is used to the research of reducing pathogenic IgG in autoimmune and alloimmune diseases.

Rozanolixizumab is a high affinity humanized immunoglobulin G4 monoclonal anti-body directed against human neonatal Fc receptor (FcRn)[1]. Rozanolixizumab binds with high affinity (dissociation constant 0.023 and 0.034nM at pH 6.0 and pH 7.4)[2]. Rozanolixizumab for 1h treatment dose-dependently inhibits human FcRn-mediated recycling of human IgG in vitro with an IC50 of 0.41nM[2]. Rozanolixizumab is an agent approved for both anti-AChR and anti-MuSK antibody-positive gMG in USA[1].
Rozanolixizumab (0.0001-100μg/mL; 4h) reduced the surface labeling of the high-affinity FcγR and FcγRI in human monocyte-derived macrophages[3].
Rozanolixizumab (30mg/kg; i.v.) for a single administration reduced the plasma IgG concentrations in cynomolgus monkeys; Rozanolixizumab (10, 30, 100 or 250mg/kg; i.v.) for a single administration rapidly decreased the plasma IgG in muFcRn−/− huFcRn Tg32 transgenic mice[2]. Rozanolixizumab (150mg/kg; from gestation day 20 until delivery; q3d; s.q.) decreased the offspring’s IgG levels at birth from pregnant female cynomolgus monkeys[4].

Preparation Method |
MDCK cells over-expressing human FcRn were plated in a 96-well plate and incubated overnight at 37℃, 5% CO2, to allow cells to adhere. The cells were incubated with Rozanolixizumab for 1h. IgG released into the supernatant was measured using a meso scale discovery (MSD) assay with an anti-human IgG capture antibody and a streptavidin-sulpho tag reveal antibody (MSD). |

Preparation Method |
