Description of Colcemid

Colcemid ist ein Zytoskelettinhibitor, der in Säugetierzellen bzw. Oozyten einen mimotischen Arrest in der G2/M-Phase bzw. einen meiotischen Arrest in der Vesikelrupturphase (GVBD) induziert[1][2]. Colchicin stört die Mikrotubuli-Polymerisation durch feste Bindung an Tubulindimere und verhindert die Bildung spindelförmiger Mikrotubuli durch Depolymerisation[7].

Zelltransformation wurde bei Dosen beobachtet, die nicht zytotoxisch waren und keine mitotische Hemmung der Zellen verursachten. Höhere Colcemid-Dosen(über 0.1 µg/ml) führten zu einer mitotischen Hemmung der Zellen und einem signifikanten Verlust der Koloniebildungsfähigkeit, aber nicht zu einer erhöhten Häufigkeit morphologischer Transformationen. In Kulturen, die mit 0.1 µg/ml Colcemid behandelt wurden, wurde eine 14-fache Zunahme der Zahl aneuploider Zellen mit nahezu diploidem Chromosomensatz festgestellt, wobei sowohl Chromosomenverlust als auch - zuwachs induziert wurden[4]. Colcemid war für Zellen in der G2 + M-Phase stärker zytotoxisch als für Zellen in der G1 + S-Phase und verlangsamte die Progression von G1-Zellen zu S. Die Effekte von Colcemid waren in aneuploiden B16-Melanomzellen deutlich stärker als in pseudodiploiden Ovarienzellen des Chinesischen Hamters(CHO)[5]. Colcemid fördert die UVC-induzierte Apoptose in Ovarialzellen des Chinesischen Hamsters(CHO.K1). Obwohl Colcemid die Entfernung UV-induzierter DNA-Schäden wie Photoprodukte oder Cyclobutan-Pyrimidin-Dimere nicht beeinflusste, akkumulierte Colcemid die DNA-Brüche, wenn es den Zellen nach UV-Bestrahlung zugesetzt wurde[3].

Eine mitotische lineare Akkumulation wurde durch kontinuierliche Colcemid-Infusion mit 5.82 µg/hr erreicht. Eine niedrige Colcemid-Dosis(0.582 und 1.455 µg/hr) über 14 Stunden führte nicht zur Ansammlung mitotischer Zellen, Dosen von mehr als 5.82 µg/hr Colcemid blockierten diese aber vollständig und akkumulierten nach einer 20-stündigen Infusion 25.5% der Zellen[6].

Vorbereitungsmethode

Die Behandlung von Embryozellen des Syrischen Hamsters in Kultur mit 0.01-0.1 µg/ml Colcemid über 48 Stunden führte zu einer morphologischen und neoplastischen Transformation der Zellen.
