Nesfatin-1 (30-59) (mouse, rat) is the active core fragment of nesfatin-1 which regulates food intakes [1]. Nesfatin-1 (30-59) can upregulate the mRNA expression of proopiomelanocortin and amphetamine-regulated transcript, and act via an oxytocin-POMC-α-MSH/melanocortin receptor 3/4 pathway to inhibit food intake in rats [2]. Nesfatin-1 (30-59) has been widely used to regulate anxiety and depression-like behavior as well as anhedonia in rats under normal weight conditions [3].
In vivo, Nesfatin-1 (30-59) treatment via a single intracerebroventricular injection at a dose of 0.9nmol/rat (5μl) reduced the cumulative food intake of diet-induced obese rats within 20 hours[4].

Preparation Method | Male Wistar rats weighing 280-350g were first group housed (4 rats/group) under controlled illumination (06.00-18.00h) and temperature (21-23°C). For the induction of diet-induced obesity (DIO), rats were fed a high fat diet (45% calories from fat, 35% from carbohydrates, and 20% from protein, 4.7kcal/g diet) for a period of 10 weeks. Nesfatin-1 (30-59) (0.9nmol/rat) or vehicle (5μl ddH2O) were injected i.c.v. directly at the beginning of the dark phase and the food intake microstructure was assessed within 24h. |
