Z-Gly-Pro-pNA is a chromogenic substrate that can be cleaved by prolyl endopeptidases[1]. Enzyme activity of prolyl endopeptidases was determined from the increase in absorbance at 410nm following cleavage of Z-Gly-Pro-pNA in 0.1M citrate/disodium phosphate buffer (pH 7), with the reaction carried out at 60°C[2]. Z-Gly-Pro-pNA has been widely used to characterize the kinetic parameters of the gluten-degrading prolyl endopeptidase from Thermococcus kodakarensis[3].
