Description of Luciferase

Luciferase is an enzyme capable of catalyzing bioluminescent reactions. Luciferase catalyzes the oxidative decarboxylation of the substrate luciferin to convert chemical energy into light energy, and it can be used to reflect the expression level of a target gene by being coupled with that gene. Luciferase can be used in research related to gene expression regulation and in vivo imaging[1-4].

Protocol of Luciferase

Establishment of a Sustained Bioluminescence Imaging Detection Method Using Luc@CaP Nanoparticles Based on Luciferase[1]

This protocol is adapted from research data and provided for reference only. Adjustments may be necessary based on specific experimental requirements.

1. Reagent Preparation: Prepare a Luciferase solution at a concentration of 400μg/mL; use calcium phosphate as the mineralization carrier to prepare Luc@CaP nanoparticles (Luc@CaP NPs) via biomimetic mineralization, with an optimal mineralization time of 4 hours; synthesize and prepare a working solution of the probe Cys(SEt)-Lys-CBT (CKCBT) at a concentration of 300μM or amino-luciferin (Am-luciferin) at 300μM; prepare a detection buffer containing 1mM ATP and 10mM Mg²⁺.

2. In vitro Cellular Level Detection:

(1) Culture the cell line and seed the cells in a 96-well plate (approximately 2×10⁶ cells per well).

(2) Add 400μg/mL of Luc@CaP NPs to the cells and incubate for 8 hours to achieve cellular internalization and uptake.

(3) Aspirate the excess reagents and add 300μM of CKCBT or Am-luciferin probe working solution to the wells.

(4) Use a live imaging system to perform real-time bioluminescence imaging (BLI) at specific time points (0, 1, 2, 4, 6, 7 hours, etc.), and record and analyze the dynamic changes in luminescence intensity.

3. In vivo Animal Level Detection

(1) Establish an orthotopic cancer model by injecting 5×10⁶ cancer cells into BALB/c mice.

(2) When the tumor diameter reaches about 7-8mm, administer 4.0mg/kg of Luc@CaP NPs via intratumoral (i.t.) injection.

(3) Twenty minutes after administration, inject 200μmol/kg of CKCBT or 50μmol/kg of Am-luciferin via intraperitoneal (i.p.) injection.

(4) Use a small animal imaging system to continuously record the bioluminescent signals at the tumor site over a time window of up to 120 minutes.

(1) Animal experiments must comply with animal welfare guidelines.
