Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp (trifluoroacetate salt) is a modified peptide containing fluorescence resonance energy transfer (FRET) groups. Fluorescence signals can be monitored using an excitation wavelength of 320nm and an emission wavelength of 420nm, making Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp commonly used as a fluorescent substrate for detecting protease activity[1].

Protocol of Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp (trifluoroacetate salt)

Assay of Proteasome Activity in Cell
This protocol is adapted from research data and provided for reference only. Adjustments may be necessary based on specific experimental requirements.
(1) Reagent Preparation: Dissolve the Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp lyophilized powder in dimethylformamide (DMF) to prepare a high-concentration stock solution (e.g., 5mM), then aliquot and store at -20°C.
(2) Enzyme Sample Preparation: Use purified PR3 enzyme (typically at concentrations ranging from 0.1nM to 10nM) or cell suspensions containing PR3 (e.g., activated neutrophils).
(3) PR3 enzyme Activity Detection: The reaction mixture contains 20μM Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp, 50mM HEPES, 150mM NaCl, and 0.05% Triton X-100, with a total volume of 200μL. Incubate at 37°C for 30 minutes. Monitor fluorescence intensity using a microplate spectrofluorometer. The excitation wavelength for Abz is 320nm, and the emission wavelength is 420nm.
Precautions:
(1) Free fluorescent substrates are subject to quenching. Please try to avoid light exposure to slow down fluorescence quenching.
(2) DMF may affect enzyme activity. The final concentration of DMF in the stock solution should be kept as low as possible (typically <1%).
(3) For your safety and health, please wear a lab coat and disposable gloves when operating.
