Aminooxyacetic acid is a gamma-aminobutyric acid transaminase (GABA-T) inhibitor. Aminooxyacetic acid exerts anticonvulsant effects by reducing GABA degradation and increasing brain GABA levels. Aminooxyacetic acid regulates hydrogen sulfide metabolism by inhibiting cystathionine beta-synthase and cystathionine gamma-lyase. Aminooxyacetic acid can be used for research on neurological diseases such as epilepsy and hydrogen sulfide metabolism[1-4].
In vitro, treatment of C6 glioma cells with 1-5mM Aminooxyacetic acid for 1-24 hours reduced intracellular ATP levels. Aminooxyacetic acid reduced glycolytic rate and extracellular lactate and pyruvate levels. Aminooxyacetic acid increased the proportion of cells in G0/G1 phase arrest. Aminooxyacetic acid reduced the proportion of cells in S phase and G2/M phase. Aminooxyacetic acid increased the proportion of early apoptotic and necrotic cells[5]. Treatment of ethanol-exposed SH-SY5Y cells with 0.5mM Aminooxyacetic acid for 12 hours reduced intracellular calcium concentration. Aminooxyacetic acid restored mitochondrial membrane potential and weakened BIP and Sec61a protein binding[6]. Treatment of MDA-MB-231 NQO1+, MDA-MB-231 NQO1-, MDA-MB-468 and T47D breast cancer cells with 100μM Aminooxyacetic acid for 2 hours reduced intracellular enrichment of alanine, citrate, glutamate and succinate[7].
In vivo, intraperitoneal injection of 10mg/kg Aminooxyacetic acid to C57BL/6J mice for 3 days after myocardial infarction surgery increased left ventricular ejection fraction and fractional shortening. Aminooxyacetic acid reduced infarct size. Aminooxyacetic acid reduced M1 macrophage proportion and increased M2-like phenotype proportion in ischemic myocardium[8]. Intraperitoneal injection of 5mg/kg Aminooxyacetic acid to acute colitis C57BL/6 mouse model for 5 days alleviated body weight loss and reduced disease activity index. Aminooxyacetic acid attenuated colon shortening and splenomegaly. Aminooxyacetic acid reduced mucosal and submucosal inflammatory cell infiltration and maintained colonic mucosal integrity[9]. Subcutaneous injection of 2mg/kg Aminooxyacetic acid every 6 hours for 24 hours (total dose 10mg/kg) to Nu/nu BALB/C mice subcutaneously implanted with HCT116 cells inhibited liver CBS activity[10].

Preparation Method |
C6 glioma cells were maintained in DMEM containing 4500mg/L D-glucose, 584mg/L L-glutamine and 10% FBS at 37°C, 5% CO2. C6 glioma cells were treated with Aminooxyacetic acid at 1-5mM for 1h, 3h, 5h or 24h. After treatment, intracellular LDH assay, Annexin V/7-AAD flow cytometry, cell cycle analysis, ATP bioluminescence assay, JC-1 assay, 2-NBDG uptake assay, extracellular lactate and pyruvate spectrophotometric assay were performed. |

Applications |
Aminooxyacetic acid decreased intracellular ATP levels at 3h and 5h. Aminooxyacetic acid increased G0/G1 phase arrest and decreased S and G2/M phase proportions. Aminooxyacetic acid reduced F4/80+iNOS+ M1-like macrophage proportion and increased F4/80+Arg1+ M2-like macrophage proportion in infarct border zone at day 3. Aminooxyacetic acid reduced iNOS and Arg1 protein and mRNA levels in heart tissue. Aminooxyacetic acid reduced H&E inflammatory cell infiltration and TNF-α positive cells, and increased CD206 positive cells at day 3.
