3,3-Dimethyl-1-butanol is an oral inhibitor that inhibits the production of trimethylamine (TMA) by microorganisms and suppresses different microbial TMA lyases[1]. 3,3-Dimethyl-1-butanol can reduce superoxide production in aorta and inhibit the occurrence of atherosclerosis and endothelial dysfunction[2]. 3,3-Dimethyl-1-butanol has been widely used to reduce TMA and gut microbiota-derived metabolite trimethylamine N-oxide (TMAO) formation in animal models[3].
In vitro, 3,3-Dimethyl-1-butanol treatment at 1μM for 24h inhibited secretion of IL-1β and IL-6 from 10µg/µl lipopolysaccharide (LPS)-stimulated mouse bone marrow-derived macrophages[4].
In vivo, 3,3-Dimethyl-1-butanol treatment (1.0% dissolved in drinking water) via oral administration for 8 weeks significantly prevented the impairment of cardiac contraction and relaxation function in mice caused by a high-fat diet[5]. Drinking 1.0% 3,3-Dimethyl-1-butanol water for six consecutive weeks can significantly reduce the plasma TMAO levels in mice undergoing aortic banding surgery, and inhibit adverse cardiac structural remodeling[6]. The continuous consumption of water containing 1% 3,3-Dimethyl-1-butanol for 21 consecutive days led to a significant reduction in the inflammatory response and oxidative stress in the limbs of mice with unilateral hindlimb ischemia, and improved perfusion recovery[7].

Cell lines | Mouse bone marrow-derived macrophages |
Preparation Method | Bone marrow was flushed and processed to a single cell suspension from the femur and tibia of 6–10-week-old DBA/1j mice using 1ml cold PBS. The single cell suspension was cultured in 9ml of complete RPMI 1640 medium supplemented with 10% fetal bovine serum, 2% HEPES, 0.6% penicillin/streptomycin, 0.1% 2-mercaptoethanol, and 20ng/ml recombinant mouse GM-CSF. After 72h, the culture media were removed and replaced with fresh differentiation media to remove non-adherent cells. After 6 days of culture, the adherent macrophages were washed with cold PBS and resuspended in RPMI 1640 without recombinant mouse GM-CSF. Cells were stimulated with 10µg/µl LPS and treated with different concentrations of 3,3-Dimethyl-1-butanol (0.00001, 0.001, 1, 100, 10000, and 25000nM) for 24h. The secretion levels of proinflammatory cytokines were analyzed. |
Reaction Conditions | 0.00001, 0.001, 1, 10, 1000, 10000, and 25000nM; 24h |
Applications | 3,3-Dimethyl-1-butanol treatment inhibited secretion of IL-1β and IL-6 from LPS-stimulated macrophages in a dose-dependent manner. |
| Animal experiment [2]: | |
Animal models | Male CD1 mice |
Preparation Method | Eight-week-old male CD1 mice were raised under a 12-hour light/12-hour dark cycle, with free access to food and water. Forty mice were selected and fed either a normal diet (ND, control group) or a high-fat diet (WD). Additionally, 1.0% 3,3-Dimethyl-1-butanol was added to the drinking water for 8 weeks (n=10 in each group). After 8 weeks of baseline and high-fat diet feeding, cardiac function was evaluated. |
Dosage form | 1.0% in drinking water for 8 weeks; p.o. |
Applications | 3,3-Dimethyl-1-butanol treatment significantly prevented the impairment of cardiac contraction and relaxation function in mice caused by a high-fat diet. |
